[metapost] ligatures

METAFONT isn't for editing .ttfs

You should be able to do as you describe (if the license allows) by:

 - installing FontForge --- free/opensource
 - loading the .ttf into that
 - creating the characters which you wish
 - assign them to appropriate code points or the private use area
 - name them appropriately
 - regenerate the font

You should then be able to use the font in xetex or luatex after making it
available to TeX.
