The default portable setup expects to be on a writable medium. For DVD
you should remove from texmf.cnf the following:

TEXMFHOME = $TEXMFLOCAL
TEXMFVAR = $TEXMFSYSVAR
TEXMFCONFIG = $TEXMFSYSCONFIG

You don't say what gives that error (I assume tlmgr).  Can you check
if tlpkg/tlperl/libpng3.dll exists?
