Very likely; I haven't used cron on OS X for a long time, so am not familiar with its environment. I'd have thought that you would need to add /usr/texbin to PATH also, but maybe tlmgr builds its own paths based on its location.

FWIW, TeX Live Utility has a feature that checks for updates and prompts you to install, but it does require interaction.
