in texmf-dist/web2c/texmf.cnf:

FC_CACHEDIR = $TEXMFSYSVAR/fonts/cache

So in an admin install, admin permissions are required to update the
font cache. Note that as of the 2016 release, the entire TL
directory is write-protected for non-admin users.

A Windows-only option in tlmgr to (re-)run fc-cache would help.

For our university installation, I have added lines

FONTCONFIG_PATH = $TEXMFVAR/fonts/conf
FC_CACHEDIR = $TEXMFVAR/fonts/cache

to <root>/texmf.cnf because I do not control the system fonts of
users.

Is there a way to run fc-cache separately for TL fonts and for
system fonts?
